easYmer® make your own custom HLA Monomers and Tetramers

More that 40 HLA allotypes have been formulated as easYmers® – hence, you can build a HLA repository of “empty” MHC class I molecules with relevance to your projects in your own lab and make custom MHC (HLA) class I monomers from day-to-day with peptides of interest. All MHC class I easYmer® molecules are biotinylated allowing you to make tetramers with your preferred labels. MHC class I easYmers® are stable in its peptide receptive form in the freezer for 24 months – in complex with peptide antigens the easYmers® are stable for years in the freezer as monomers.

To ensure optimal staining of your CD8+ T cells, we strongly recommend to experimentally determine if the peptide of interest supports peptide MHC (pMHC) complex formation The easYmer kit includes a simple FACS based protocol allowing you to validate proper pMHC class I interaction (click here for protocol).

For optimal HLA tetramer staining of CD8+ T cells, please follow our MHC class I tetramer staining protocol.

We also recommend that you use predictors (e.g. NetMHC series or NetMHCpan) to assist you in your design of suitable monomers.

The resulting HLA monomers can easily be tetramerized with labelled streptavidin (e.g. fluorophore label for analysis in a classical flow cytometry assay or metal-label for analysis by CyTOF). The pMHC monomers can also be long-term stored frozen for later tetramerization.

The easYmer technology is highly flexibility and suitable for:

  • Evaluation of peptide-MHC class I interaction.
  • Epitope / Neo-epitope screening – small-scale tetramer production for screening of a large number of different epitopes in parallel (i.e. for staining of up to five T cell samples per HLA tetramer).
  • Donor screening – full-scale tetramer production for screening of a single epitope in a large number of samples (i.e. for staining of up to 50 T cell samples per HLA tetramer).
  • 30+ HLA-A | HLA-B | HLA-C allotypes available including HLAs A*01 | A*02 | A*03
